S-Adenosyl-L-Methionine
Drives 100+ methylation reactions including neurotransmitter synthesis — clinically validated for depression, osteoarthritis, and liver health.
What the evidence says
Most SAMe studies are mechanism or observational rather than RCTs that measure a clinical effect — keep findings provisional.
Most evidence is from high-quality meta-analyses and randomised trials published 2004–2026 with a typical study size of 656 participants.

SAMe has an evidence score of 9/10 — very strong evidence based on 14 indexed studies, including 32 meta-analyses. Drives 100+ methylation reactions including neurotransmitter synthesis — clinically validated for depression, osteoarthritis, and liver health.
The commonly studied dose of SAMe is 400-1600mg for mood; 600-1200mg for joints. Research points to an estimated optimal dose around 1000mg, with a minimum effective dose near 600mg. Individual response varies — start low and adjust.
The best time to take SAMe is in the morning. It can be taken on an empty stomach. S-adenosylmethionine (SAMe) is the body's primary methyl donor, involved in >100 methylation reactions including neurotransmitter synthesis, DNA methylation, and phospholipid metabolism.

SAMe is a molecule naturally produced in the body that's involved in over 100 biochemical reactions including neurotransmitter synthesis, joint cartilage maintenance, and liver detoxification. Clinical evidence supports its use for depression (comparable to some antidepressants), osteoarthritis, and liver conditions. One of the most clinically validated supplements.
Provides methyl groups for critical reactions
Supports synthesis of mood-regulating neurotransmitters

Minimum: 4 weeks
Optimal: 12 weeks
Cycling: Not required
Note: Best absorbed on empty stomach. Avoid evening dosing as it may be energizing. Can take with B12 and folate to support methylation.
Meta-analysis of 656 patients showed small effect size (SMD -0.17) corresponding to only 0.4cm improvement on 10cm pain scale. Effect is modest and barely reaches statistical significance threshold.
Significant antidepressant effects
Reduced osteoarthritis symptoms
Improved liver function in liver disease
Insufficient data; avoid
Avoid — may trigger manic episodes
Risk of serotonin syndrome — use only under medical supervision
May reduce effectiveness
Increased serotonin syndrome risk
Serotonin syndrome risk
Hypertensive crisis, serotonin syndrome
Tip: Start low, use enteric-coated, take with small snack if needed
Tip: Reduce dose
Tip: Take earlier in day
Tip: Contraindicated in bipolar disorder
SAMe is generally safe at recommended doses, with a few precautions worth noting. The most commonly reported side effects are GI upset/nausea, anxiety/restlessness, insomnia. Use caution if any of these apply to you: Bipolar disorder (may trigger mania); Parkinson's disease on levodopa.
