The 300 mg point rests on four 12-week placebo-controlled trials of standardised extracts at 300 mg a day. Three found better verbal learning or delayed recall than placebo (PMID 11498727, 20590480 with 98 older adults, 18611150 with 54 adults over 65). The largest, 101 adults with memory complaints (PMID 41091332), found no difference in verbal learning, attention or working memory. The 160 mg point is one 2-month trial in 62 people with mild cognitive impairment (PMID 38538390): attention improved, and the overall cognitive score differed from placebo only at the final visit. No trial compared doses within one study, and none gave more than 300 mg a day for weeks, so no optimal dose or safety ceiling can be read from them. Two trials reported more gastrointestinal complaints on bacopa.
