D-Chiro-Inositol
An inositol isomer used for PCOS and insulin sensitivity — but the strong evidence is for myo-inositol or the myo+D-chiro COMBINATION; D-chiro-inositol ALONE (especially at high doses) is less proven and may even impair oocyte quality.
What the evidence says
Most D-Chiro-Inositol studies are mechanism or observational rather than RCTs that measure a clinical effect — keep findings provisional.
Most evidence is from medium-quality meta-analyses and randomised trials published 2017–2024.

D-Chiro-Inositol has an evidence score of 4.2/10 — emerging evidence based on 4 indexed studies, including 2 meta-analyses. An inositol isomer used for PCOS and insulin sensitivity — but the strong evidence is for myo-inositol or the myo+D-chiro COMBINATION; D-chiro-inositol ALONE (especially at high doses) is less proven and may even impair oocyte quality. Representative study: PMID 38163998.
The commonly studied dose of D-Chiro-Inositol is Best used as myo-inositol:D-chiro-inositol 40:1 (e.g. 2000mg MI : 50mg DCI); DCI alone is not recommended at high doses. Individual needs vary — start at the lower end of the range and adjust based on how you respond.

D-chiro-inositol (DCI) is one of the inositol stereoisomers involved in insulin signaling. It's marketed for PCOS, insulin sensitivity, and ovulation. The honest picture: most positive evidence is for myo-inositol (MI) or the MI+DCI combination at a physiological ~40:1 ratio — DCI ALONE, particularly at high doses, is less supported and research suggests excess DCI can actually impair oocyte quality and the MI:DCI balance. So DCI is best understood as a component of inositol therapy rather than a standalone high-dose supplement. (See myo-inositol for the better-evidenced primary form.)
DCI is an inositol-phosphoglycan mediator of insulin action, improving insulin sensitivity.
Modulates ovarian insulin signaling; the MI:DCI ratio matters for oocyte quality.

Take with food
| Form | Type |

Optimal: 12 weeks
Cycling: Not required
Note: With food; within an MI combination.
Dose-response data unavailable. The current published research for D-Chiro-Inositol does not provide sufficient dose-specific outcome data to generate reliable dose-response curves.
Refer to the Dosage & Timing section above for recommended dose ranges based on available evidence.
Supports ovulatory cycles in PCOS (best in MI:DCI combination).
Improves insulin sensitivity.
High-dose DCI alone may impair the MI:DCI balance / oocyte quality.
Avoid high-dose DCI alone — may impair oocyte quality; use MI-dominant blend.
Monitor blood glucose.
Inositols improve insulin sensitivity — monitor for additive glucose-lowering.
Tip: Take with food
The best time to take D-Chiro-Inositol is with meals. Take it with food. Taken with meals; use within a myo-inositol combination rather than high-dose DCI alone.
D-Chiro-Inositol is generally safe at recommended doses, with a few precautions worth noting. The most commonly reported side effects are mild GI upset. Use caution if any of these apply to you: Pregnancy (use only under guidance); Use with antidiabetic drugs (additive).
Activates AMPK to regulate blood sugar, improve insulin sensitivity, and support lipid metabolism — comparable to metformin in some trials.
