Effect of glucomannan on plasma lipid and glucose concentrations, body weight, and blood pressure: systematic review and meta-analysis
The American journal of clinical nutrition (2008) · Meta analysis · n=531
Meta-analysis of 14 randomised controlled trials with 531 participants.
The Impact of Glucomannan, Inulin, and Psyllium Supplementation (Soloways(TM)) on Weight Loss in Adults with FTO, LEP, LEPR, and MC4R Polymorphisms: A Randomized, Double-Blind, Placebo-Controlled Trial
Nutrients (2024) · Rct · n=112
The experimental group showed significant reductions in body weight (treatment difference: -4.9%; 95% CI: -6.9% to -2.9%; p < 0.01) and BMI (treatment difference: -1.4 kg/m2; 95% CI: -1.7 to -1.2; p < 0.01) compared to placebo.
Comparative effects of nutraceuticals on body weight in adults with overweight or obesity: A systematic review and network meta-analysis of 111 randomized clinical trials
Pharmacological research (2023) · Meta analysis · n=6171
We conducted random-effects network meta-analysis with a Frequentist framework to estimate mean difference [MD] and 95% confidence interval [CI] of the effect of nutraceuticals on weight loss.
Effect of 12-Week Intake of Nicotinamide Mononucleotide on Sleep Quality, Fatigue, and Physical Performance in Older Japanese Adults: A Randomized, Double-Blind Placebo-Controlled Study
Nutrients (2022) · Rct · n=108
Significant interactions were observed between 5-STS and drowsiness. 5-STS of all groups on post-intervention and drowsiness of the NMN_PM and Placebo_PM groups on mid- and post-intervention showed significant improvement compared with those in pre-intervention.
Anti-inflammatory effects of nicotinamide mononucleotide (NMN) in human skeletal muscle after BFR-exercise
Journal of the International Society of Sports Nutrition (2026) · Rct · n=11
Randomised placebo-controlled crossover trial in 11 untrained young men taking NMN 1,200 mg a day for 7 days before blood flow restriction resistance exercise, with muscle biopsies.
The differential impact of three different NAD(+) boosters on circulatory NAD and microbial metabolism in humans
Nature metabolism (2026) · Rct · n=65
Ultimately, we propose a gut-dependent model for the modes of action of the three NAD+ precursors with NR and NMN elevating circulatory NAD+ via the Preiss-Handler pathway, while rapidly absorbed Nam acutely affects NAD+ levels via the salvage pathway.
Two trials of glucomannan on its own report both a dose and a weight outcome. In a single-blind, placebo-controlled crossover of 20 adults with type 2 diabetes, 3 g a day for 4 weeks lowered BMI, a secondary result (PMID 18041436). In a randomised placebo-controlled trial of 53 overweight or moderately obese adults, 1.33 g before each of three meals (about 4 g a day) for 8 weeks did not change weight compared with placebo, and gastrointestinal symptoms, liver enzymes and creatinine did not differ from placebo (PMID 24490058). The curve stays flat at 4 g for that reason. No trial compared doses. The weight meta-analyses cited here do not state a dose, so they are not plotted.
The 300, 600 and 900 mg points come from a 60-day dose-ranging trial in 80 healthy middle-aged adults (PMID 36482258): every dose raised blood NAD against placebo, with the highest levels at 600 and 900 mg, and no safety issues up to 900 mg. The 250 mg point is a 12-week placebo-controlled trial in 60 older adults where blood NAD+ rose (PMID 38789831). A pharmaceutical formulation, MIB-626, gave dose-related NAD increases at 1,000 and 2,000 mg a day for 14 days (PMID 35182418) and is not plotted because it is a different formulation. A higher blood NAD level has not been shown to translate into clinical benefit.
